1. First, remove the inner sterilization bucket, and then add an appropriate amount of water to the outer pot to make the water surface level with the triangular shelf.
2. Put it back into the sterilization bucket and fill it with the items to be sterilized. Be careful not to pack too tightly to avoid obstructing steam circulation and affecting sterilization effectiveness. The triangular flask and the end of the test tube should not come into contact with the barrel wall to prevent condensation water from wetting the paper at the mouth and penetrating into the cotton stopper.
3. Cover and insert the covered exhaust hose into the exhaust slot of the inner sterilization bucket. Tighten the two opposite bolts symmetrically in pairs to ensure consistent tightness and prevent air leakage.
4. Heat the pot with an electric stove or gas, and open the exhaust valve at the same time to boil the water and remove the cold air inside the pot. After the cold air is completely exhausted, close the exhaust valve and let the temperature inside the pot gradually rise with the increase of steam pressure. When the pressure inside the pot reaches the desired level, control the heat source and maintain the pressure for the required time. This experiment uses 1.05kg/cm2121.3 ℃ for sterilization for 20 minutes.
5. After the sterilization time is up, cut off the power or turn off the gas to allow the temperature inside the sterilization pot to naturally decrease. When the pressure on the pressure gauge drops to 0, open the exhaust valve, loosen the bolt, open the lid, and remove the sterilized item. If the pressure is not reduced to 0 and the exhaust valve is opened, the sudden drop in pressure inside the pot will cause the culture medium in the container to flush out of the flask or test tube due to the imbalance of internal and external pressure, resulting in contamination of the cotton plug with the culture medium.
6. Place the sterilized culture medium in a 37 ℃ incubator for 24 hours. If there is no growth of miscellaneous bacteria after inspection, it can be used for later use. However, there are a few points to note:
1. There must be sufficient water in the pot.
The airtightness of the pot must be very good.
3. After use, be sure to place it at a suitable temperature before opening it.
